A 40-year-old Princes Hill man died in hospital a day after he was struck by the car on Moreland Road, Coburg, on the evening of August 24.
Police arrested the woman in Lower Templestowe on Monday, alleging she was the driver.
She has been charged with failing to stop at the scene of an accident, failing to render assistance, failing to report a collision to the police, and driving while suspended.

A 61-year-old man was also arrested in Abbotsford yesterday in relation to the same incident, but he has since been released without charge, pending further inquiries.
Following the man’s death, police released footage of a white 2016 Audi Q5 SUV believed to be the car involved in the collision.
Police are still searching for that vehicle, which they say will likely have visible damage to both the front and passenger side.

The investigation remains ongoing, with detectives still seeking footage and witnesses to the crash. Anyone who can help is urged to contact Crime Stoppers.
The charges come as the group Safe Moreland Rd Now seeks signatures for their petition calling on the state government to implement reduced speed limits, protected bike lanes, and more signalised pedestrian crossings throughout the length of Moreland Road.
The organisers say their action has been prompted by “recent tragic deaths and numerous near misses along Moreland Road, through Brunswick West, Coburg and Brunswick East”, including this one.
In an unrelated incident on Sunday night, in the southern Grampians town of Dunkeld, a cyclist was killed when her bike collided with a Toyota Landcruiser.
Also on Sunday, a 20-year-old man was charged after allegedly hitting a cyclist in the outer western Melbourne suburb of Wyndham Vale on January 24. That cyclist died at the scene.
